The current clinical classification of multiple sclerosis (MS) based on the brain lesions assessed by magnetic resonance imaging (MRI) is pathologic, prone to inter and intra-examiner variability, and is costly. We scanned a cohort of 21 MS patients (9 in relapse, 12 in remission) and 19 healthy controls (HC) with functional MRI (fMRI) while performing a verbal working memory task. We classified the patients and HC into five subgroups based on the main anatomical and functional patterns identified in each subject: HC-1: HC; HC-2: HC with resting-state spatial coherence; MS-1: MS with resting-state spatial coherence; MS-2: MS with abnormal functional connectivity; MS-3: MS with abnormal functional connectivity and abnormal spatial coherence; MS-4: MS with abnormal functional connectivity, spatial coherence and both increased functional connectivity and functional integration.
